COXI sequencing

Sir,I have got a curated specimen of Viverridae family. The specimen is dried and kept in the museum for many years. I doubt that it is treated with borric powder and other preservative used in taxidermy. How can we remove the inhibitant from the sample of tissue for the purpose of sequencing. Is there any common or specific primers available for this group.See More

"Hi Sreejith, Try to align your sequences with sequences without frame shift and find out the position where problem lies. Then try to compare with .ab files. Which software do you use for editing trace files"

Gaikwad Swapnil Sopan replied to sreejith kanholi's discussion 'Sequence error' in the group Problems in DNA barcoding
"Dear Sreejith This problem is common if your trace file is not properly corrected, sometimes unknowingly bases are inserted. This leads to frame shift. correct your trace files once again Regards Dr. Swapnil Gaikwad"

Sequence error

I have doing my deposition of sequence in the NCBI, but when one of the sequence is deposited I have got a message stating that some or all of the sequences contain reading frame shifts (insertions/deletions based on BLAST similarity search results and/or an alignment). How can we check it is there any software available. See More
