If you are using universal primer, sometimes it doesn't give amplification for few samples. This is my personal experience. Try do design more specific primer for mollusc.

Finally, this method didn't work with the species that I work with... I don't understand! I know that my extractions are good, its PCR that didn't work.

I dont which primer you are using but Folmer LCO and HCO usually gives application at 47 degrees, try this.

I amplified at 51 degrees (anealing temperature). After that, try to use a program proposed by D. Steinke (anealing temperature : touch up 40-44) that ordinary work well with gastropods but I amplified nothing... Does anyone work with mollusc barcoding?Thank you very much!GenevièveSee More

- I'll make DNA barcode sequences template of several species that we collected in our cages (monitoring tool to detect hotspot biodiversity in the bay of fundy). I'll also try to determine if an important biodiversity will interfer with benthic community biotic resistance. The ascidians Ciona intestinalis is the invasive species of interest. Finally, I'll use barcoding to determine the presence of potential cryptic species of the genus Ciona in this area.
