In an attempt to define a barcode for Bacteria, (a continuation from previous discussion entitled "Bacterial DNA barcoding")I did a comparison of rpob and 16S rRNA in Pseudomonas spp. I have selected Pseudomonas as the genus has higher number of species recorded so far. I compared rpoB and 16S rRNA gene sequences of 20strains of Pseudomonas fluorescence. rpoB of this strain contained approx. 10times more variation than its 16s rRNA gene sequence. Please find the enclosed phylograms (of 16s rRNa nad rpoB) for 20strains of P.fluorescence. One could agree that rpoB has high resolution power than 16S rRNA. I have used 1100bps (averagely) of rpob and 1300bps of 16s rRNA (averagely) for phylogram construction. I am about to analyse the rpoB sequences of 100spp of Pseudomonas. I will let the forum know the results very soon.
