I would appreciate member's comments on preserving adult mosquitoes for DNA barcoding. Mosquito larvae in ethanol are OK, but adults?? very fragile and small.

Comment by Matthew Bowser on January 7, 2011 at 1:27pm Monitoring of Culicidae and other terrestrial arthropod taxa using next-gen (environmental sampling) DNA barcoding methods is the long-term objective of a mini-bioblitz we are planning for this summer. This field season, we will be sampling Culicidae as well as other groups, obtaining determinations on voucher specimens (for which I am seeking collaboration), and submitting these specimens to a DNA barcoding lab to build a regional DNA barcode library.
Comment by Richard Wilkerson on January 7, 2011 at 11:47am Are you looking for collection and archiving methods? Basic taxonomy to anchor your sequences is quite important for validating your results. If so, I can point you to a summary of field and laboratory methods.
Do you have a "nex-gen" method on the horizon? I too am interested in this, especially for Anopheles.

This is part of a larger arthropod inventory initiative on therefuge, but the Culicidae is a priority group because of their abundance, ecologicalimportance, and prevalence in samples we obtained for our long-term monitoringefforts. We are building a referencelibrary of DNA barcodes for the refuge to set the stage for monitoring usingnext-generation DNA barcoding methods.
