Preserving mosquitoes for DNA barcoding 1 Reply

I would appreciate member's comments on preserving adult mosquitoes for DNA barcoding. Mosquito larvae in ethanol are OK, but adults?? very fragile and small.

Monitoring of Culicidae and other terrestrial arthropod taxa using next-gen (environmental sampling) DNA barcoding methods is the long-term objective of a mini-bioblitz we are planning for this summer.  This field season, we will be sampling Culicidae as well as other groups, obtaining determinations on voucher specimens (for which I am seeking collaboration), and submitting these specimens to a DNA barcoding lab to build a regional DNA barcode library.

Are you looking for collection and archiving methods?  Basic taxonomy to anchor your sequences is quite important for validating your results.  If so, I can point you to a summary of field and laboratory methods.

Do you have a "nex-gen" method on the horizon? I too am interested in this, especially for Anopheles.

I am looking for help with inventorying and obtaining DNAbarcodes of mosquitoes from the Kenai National Wildlife Refuge thissummer.  This is part of a larger arthropod inventory initiative on therefuge, but the Culicidae is a priority group because of their abundance, ecologicalimportance, and prevalence in samples we obtained for our long-term monitoringefforts.  We are building a referencelibrary of DNA barcodes for the refuge to set the stage for monitoring usingnext-generation DNA barcoding methods.

On the opening day of the Third international Barcoding Conference, Mexico, Dr Yvonne Linton, on behalf of the MBI consortium presented barcode data for 1900 specimens, corresponding to some 174 recognised mosquito species. Significantly, some 46 cryptic taxa were additionally recovered using DNA barcodes, indicating that mosquito biodiversity make currently be under-represented by some 26%. Introgression, as evidenced by additional ITS2 sequencing was noted in four species pairs. Total data presented included 35% of known species of Anopheles, which includes 90% of reported malaria vectors world-wide.

In addition, Dr Pradeep Kumar gave an excellent presentation on Indian species, to add to his previously published paper on barcoding Indian taxa (link to Kumar et al., 2007) - we invite Dr Kumar to give us an summary of his data.Kumar et al. 2007.pdf
