Hi all
I am about to go on an official barcoding trip in South Africa. Besides the small ascomycetes that is more my specialty, I will also collect some larger fungi. THis trip has a public profile, so the larger fungi will be there for the people to better understand and to attract their attention. Because I want to do it properly, are there any do's and dont's when I collect the larger fungi and preparing them for barcoding, e.g. how much tissue must I put in directly, I should probably dry first? THere are those of you with much more experience than me. I guess I will stick to the more robust ones and leave the fragiles ones for later.
I have barcode kits.

What I have done in the past is put a small amount of fresh tissue in a buffer (e.g. CTAB) in a 1.5 mL tube, then dry down the rest of the specimen for the herbarium. You will need to have collection codes to link the samples of course. (e.g. http://twitpic.com/4xwvvu )
