Hello sir/madam,

I am currently working on  DNA Barcoding of the odonata.Am in the initial stages of sample collection and only limited referece are available on this area of study.Already, literature is available on 16s and 28s mitochondrial genes, there is no data available on the CO1 gene.So please suggest me which gene is ideal for the species identification and if any specific primer is suitable for the data mentioned above kindly suggest me,do the needful.

I think that if you are doing barcoding of insects the best possible way to start is with the partial COI gene in the mitochondria as this is officially the barcode gene for insects and other organisms. You could extract DNA and amplify using the folmer primers and see what results you get. You should get a fragment of atleast 658bp.

ya yes i get it

thank you

Have you tried it already? IS the official barcode not being used for Odonata? I assume you are talking about dragonflies etc.

Well I think the partial COI (by uce of the Folmer primers) should work. If not, that could be interesting even for me. Please let me know how this has turned out.
