Hi everybody,

My work group  and I are trying to get DNA from ancient seeds trying (Solanum,  Cucurbita, Lagenaria and Bunchosia samples) which varied in age from 50 - 350 years old or even more. Can anybody suggest us a protocol that works?. I have found somes like  phenol-based protocols but still no good results.

Im Dhivya.S, Mphil Scholar, Bharathiar University, India. I work to authenticate the Traditional Indian Herbal Medicines, which are composed of dried parts of the plant tissue...i ve send u a pdf file...from tat i ve tried few protocols to extract dna from the dried samples and found to be promising for PCR amplification...i ve also tried PVP, CI-Phenol based methods... Pls do suggest me some protocols whichever u ve tried...

I am attaching some protocols that I already tested. Good for some plants but for all of them (specially for cinchona sp. that its a Rubiaceae).

Me again, we have experience in extracting decades bones for forensic DNA. In this case, CTAB usually perform well over the guanidine hcl. Or simply use QIagen investigator kit. further, try to extract the core part of the seed and at least do some peel off for the outer. remember, extracting DNA for PCR only requires a very very little amount of sample, dont be greedy, quality is the priority.
